2) Incubation Technique :

An incubation technique is very important aspect of cultivation of Mircobes after inoculation process completion.In this technique microbes can grow and reach upto their optimum growth by supplying them their suitable & favourable temperature, humidity, & other gases conditions such as CO_{2 &} O₂ contents of atmosphere inside in a specific device called Incubator.

In simple word its a device or equipment whuch is used for the optimal growth keep the organisms or provide them sustainable suitable conditions to growth in well defined temperature and also with the help of other growth factors in Vitro.

The device used to complete the II technique in Cultivation of Microbes is known as Incubator.

An incubator is a device which can be used as to provide microbes proper growth conditions which are artificial conditions in under super vision of Microbiologists.

With the help of providing Optimum growth temperature supply, humidity & maintaining gas contents & in it like Carbon dioxide & Oxygen Promotes the multiplication of microbes in 2(n).

Its totally depends on organism where time, temprature,humidity and gas Contents varies. Where im multiplication of microbes it can take by over period of time in hours, days and in case of some organisms it can take weeks too.

With the help of process of incubation it is clear that production of culture over keeping them for specific period of time or decided or optimum time and temprature Visible growth of microbes can be observed easily by the Microbiologist.

Temperature depend terminology for Microbes :

1.Psychrophile/Cryophile

2.Mesophiles

3.Thermophiles etc.

1.Psychrophiles :

The microbes which grows on temperature range between -20°C to +10°C are refer as "Psychrophiles/Cryophiles".

2.Mesophiles : 

The microbes which growt can be done in range of between 20°C to 40°C are refer as "Mesophiles".

3.Thermophiles : 

The microbes which grow in range of between 41°C to 60°C are refer as "Thermophiles". Optimum temp is 50°-55°C.
